Description

Data is associated with the analysis of proteins (cell wall lysins and lipoproteins) involved in bacterial spore germination. The dataset is sorted into four folders, two of which contain phase contrast and fluorescence microscope images of sporulating bacterial cells and spores. The third folder contains Excel spreadsheets with germination kinetic data for various strains of bacterial spores. These data were generated with a microplate reader monitoring the absorbance of spore suspensions over time. Raw data was arranged into columns and in some cases averaged to facilitate generation of graphs. The final folder contains files associated with structural models of proteins of interest and files associated with bioinformatic (EVcouplings) analyses. The latter were generated using the EVcouplings server (https://evcouplings.org) and analysed using a proprietary Jupyter notebook, a link to which is included in the folder. The dataset also includes a Readme.txt file with additional information.

Software / Usage instructions

Germination kinetic data was generated in Microsoft Excel (saved as spreadsheets in .xlsx format). Microscope images were taken with QCapture software and saved as .tif files. These can be opened in standard graphics packages e.g., PhotoShop. Molecular models were generated by AlphaFold and saved as .cif or .pdb files, which can be opened and used with standard molecular graphics software packages e.g. PyMOL. EVcouplings config and output files in .txt and .csv format. A custom plotting Jupyter Notebook can be used to visualize EVCouplings outputs, a link to which is provided in the deposited Structural Models folder.
